The analysis of fatty acids profile is a fundamental detemination for the characterization of raw materials and processed foods. This determination involves the fat hydrolysis followed by the methylation of free fatty acids. Many methods have been developed in order to avoid the undesirable deteriorative changes during the procedure (oil oxidation, artifacts formation) and to minimize the time of analysis. However, each method presents some disadvantages and it was not yet established the most efficient one. Thus, the aim of this work was to compare hydrolysis and methylation methods by using methanolic solutions of 2% H2SO4, 5% HCI, 14% BF3, and 5% CH3COCI. The one-step hydrolysis/methylation method without prior lipid extraction was also tried on freeze-dried samples. The results obtained suggest a higher hydrolysis/methylation yield obtained with the CH3COCI/CH3OH system, no artifacts formation with BF3/CH3OH was detected and the utilization of freeze-dried samples seemed to be an interesting way for the analysis of fish fatty acids.
